PRODUCT OVERVIEW

PRMT1 is a recombinant type I protein arginine methyltransferase expressed as a fusion protein in bacteria.  As supplied, the enzyme is active and capable of methylating arginine acceptor sites in peptides and proteins under appropriate incubation conditions.

PURIFICATION

Glutathione Sepharose chromatography.

FORMULATION

Tris Buffer pH 8.

RECOMMENDED USAGE                              

In vitro methylation of proteins and peptides.  A pilot assay is suggested to determine the amount of enzyme appropriate for the end user’s assay conditions.  CH3 BioSystems has used 5 ml of this lot for detection of radioactive methyl incorporation in a 24 ml assay volume and 15 ml of this lot for non-radioactive detection using Anti-mRG in a 30 ml assay volume.

TESTING

All lots of PRMT1 exhibit methyltransferase activity based on the conversion of an arginine-rich peptide to a methylarginine containing peptide based on methylarginine-specific antibody reactivity.  Arbitrary activity units based on a standardized proprietary formula.

STORAGE

Avoid multiple freeze/thaw cycles.  Thaw upon first use and prepare appropriately sized aliquots.  Store at -80°.  Stable for 6 months from date printed on label.
